The brain structure and genetic mechanisms underlying the nonlinear association between sleep duration, cognition and mental health. Nature Aging, 2022; DOI: 10.1038/s43587-022-00210-2

Sleep plays an important role in enabling cognitive function and maintaining good psychological health. It also helps keep the brain healthy by removing waste products. As we get older, we often see alterations in our sleep patterns, including difficulty falling asleep and staying asleep, and decreased quantity and quality of sleep. It is thought that these sleep disturbances may contribute to cognitive decline and psychiatric disorders in the aging population.

In research published today in Nature Aging, scientists from the UK and China examined data from nearly 500,000 adults aged 38-73 years from the UK Biobank. Participants were asked about their sleeping patterns, mental health and wellbeing, and took part in a series of cognitive tests. Brain imaging and genetic data were available for almost 40,000 of the study participants.

By analysing these data, the team found that both insufficient and excessive sleep duration were associated with impaired cognitive performance, such as processing speed, visual attention, memory and problem-solving skills. Seven hours of sleep per night was the optimal amount of sleep for cognitive performance, but also for good mental health, with people experiencing more symptoms of anxiety and depression and worse overall wellbeing if they reported sleeping for longer or shorter durations.

The researchers say one possible reason for the association between insufficient sleep and cognitive decline may be due to the disruption of slow-wave ? ?deep? ? sleep. Disruption to this type of sleep has been shown to have a close link with memory consolidation as well as the build-up of amyloid ? a key protein which, when it misfolds, can cause ?tangles? in the brain characteristic of some forms of dementia. Additionally, lack of sleep may hamper the brain?s ability to rid itself of toxins.

The team also found a link between the amount of sleep and differences in the structure of brain regions involved in cognitive processing and memory, again with greater changes associated with greater than or less than seven hours of sleep.

Having a consistent seven hours? sleep each night, without too much fluctuation in duration, was also important to cognitive performance and good mental health and wellbeing. Previous studies have also shown that interrupted sleep patterns are associated with increased inflammation, indicating a susceptibility to age-related diseases in older people.

Professor Jianfeng Feng from Fudan University in China said: ?While we can?t say conclusively that too little or too much sleep causes cognitive problems, our analysis looking at individuals over a longer period of time appears to support this idea. But the reasons why older people have poorer sleep appear to be complex, influenced by a combination of our genetic makeup and the structure of our brains.?

The researchers say the findings suggest that insufficient or excessive sleep duration may be a risk factor for cognitive decline in ageing. This is supported by previous studies that have reported a link between sleep duration and the risk of developing Alzheimer?s disease and dementia, in which cognitive decline is a hallmark symptom.

Professor Barbara Sahakian from the Department of Psychiatry at the University of Cambridge, one of the study?s authors, said: ?Getting a good night?s sleep is important at all stages of life, but particularly as we age. Finding ways to improve sleep for older people could be crucial to helping them maintain good mental health and wellbeing and avoiding cognitive decline, particularly for patients with psychiatric disorders and dementias.?

The research was supported by the National Key R&D Program of China, the Shanghai Municipal Science and Technology Major Project, the Shanghai Center for Brain Science and Brain-Inspired Technology, the 111 Project, the National Natural Sciences Foundation of China and the Shanghai Rising Star Program.

Indian-American actor and director M. Night Shyamalan has made a name for himself in the filmmaking world. As his movies manage to be hits and blockbusters, they garner praise for how unique their storylines are, as well as how they manage to twist in on themselves and captivate audiences even when it seems like they have run out of steam. He entered the movie scene in 1992 with Praying With Anger, a film he starred in, wrote, and produced. His first two movies were financed by his parents and friends, while, as he revealed later (via EW), he was a ghostwriter for She?s All That.

However, Shyamalan has established a name for himself since then and has released a plethora of successful and unsuccessful movies. Soon, he will be directing Knock at the Cabin, a movie that features the likes of Dave Bautista, Harry Potter?s Rupert Grint, and Hamilton?s Jonathan Groff. Until then, fans can enjoy these movies, which are some of the best of Shyamalan?s filmography.

7 After Earth

Before he was an Academy Award-winning actor, Will Smith created the story for After Earth and starred in it with his son, Jaden Smith, alongside Zoë Kravitz. Many centuries later, Earth has become uninhabitable, and humans live in space colonies. They are at war with aliens known as the S?krell, and the human brigade called The Ranger Corps is in charge of taking out any threats. Cypher (Will Smith), a general at Ranger Corps, has a son who wants to become one himself, but keeps failing. When the father-and-son duo crash lands on Earth, they find themselves facing threats unlike anything they have seen before. After Earth lacks any real pacing, making it difficult to watch, and leans towards tendencies where one lectures about morals instead of anything happening.

6 The Visit

The Visit marked Shyamalan?s return to the horror genre in 2015, and it did not disappoint in his classic style. Two teenage siblings live with their divorced mother in Philadelphia, who has not spoken to her parents in fifteen years. One day, the siblings? grandparents track them down online and invite them to stay for five days at their rural farmhouse. The family agrees, but when the children arrive at the house, their grandparents begin to act strangely. The Visit is unsettling and vaguely reminiscent of the gory drama sealed inside original, unedited fairy tales. It is a layered nightmare, a fresh dash of originality in Hollywood horror movies.

5 Unbreakable

Released in 2000, Unbreakable marks the beginning of the Unbreakable series that includes Shyamalan?s later films Glass and Split. A former star quarterback turned security guard boards a train, which then crashes. When he wakes up in the hospital, he is not injured when there was no way he could have survived that. He begins realizing that he has never been sick and that he shows paranormal capabilities when it comes to strength, thus making people believe he is indeed a superhero. Unbreakable is now considered one of the best superhero films ever released, helping distinguish it from others.

4 Old

Before one?s beach vacation, they better not watch Shyamalan?s latest movie Old. Gael García Bernal and Vicky Krieps star as a couple preparing to finalize their divorce. To ease their young children’s anxieties on the divorce, they agree on one last family vacation together to a tropical resort. The hotel’s employees take the family to a private beach with a couple of other parties, but the beach seems to have supernatural qualities as its occupants are rapidly aging. With its musings on time and how it passes too quickly, alongside medical exploitation, Old?s twist offers lessons in keeping an audience?s attention.

3 Split

Split is the second movie in the Unbreakable series, and it stars James McAvoy, Anya Taylor-Joy, and Betty Buckley. A man (McAvoy) struggling with dissociative personality disorder has 23 personalities. One night, his personality named Dennis takes over and kidnaps three girls outside a birthday party. They attempt to escape from him before his 24th personality, named ?The Beast,? takes over and kills them all. McAvoy is the standout from this film for his performance as the young man with split personalities.

2 Signs

2002?s Signs starred Mel Gibson and Joaquin Phoenix. A former priest (Gibson) has renounced his faith in God and lives on a farm with his two children and younger brother (Phoenix). One day, crop circles begin to appear around the farm, but they make nothing of it. When crop circles and flashing lights begin to appear all over North America, it has become obvious to the ex-priest that this could only be the work of aliens. Although the movie does tend to directly deliver information, its ambiance makes up for what it lacks.

1 The Sixth Sense

The Sixth Sense is M. Night Shyamalan?s best film to date, and it was the movie that introduced his work to a larger audience. A child psychologist (Bruce Willis) discovers one of his clients, a nine-year-old (Haley Joel Osment) is capable of speaking to the dead and can see ghosts wandering around. The Sixth Sense is partially a ghost story and a horror movie, but that does not stop it from making it a really good movie. Osment?s performance was particularly praised, as well as the movie?s script.

Since the first successful fabrication of a two-dimensional structure of carbon atoms about 20 years ago, graphene has fascinated scientists. A few years ago, researchers discovered that two layers of graphene, slightly twisted against each other, can conduct electric current without loss. In recent years, this discovery has prompted scientists to explore such layered materials in greater detail. A recent notable example is mirror-symmetric twisted trilayer graphene, where three layers of graphene are stacked with alternating twist angles. It is the first moiré system that can both be efficiently tuned with a perpendicular electric field and was demonstrated experimentally to exhibit robust superconductivity, alongside various other phases. ?This establishes trilayer graphene as an exciting platform for complex many-body physics, but the nature of the observed interaction-induced insulators, semi-metals, and superconductivity remains unknown,? says Mathias Scheurer from the Department of Theoretical Physics of the University of Innsbruck.

In a paper published in Physical Review X, a team led by Scheurer numerically and analytically studied the phase diagram of this system for different numbers of electrons per moiré unit cell and as a function of electric field. ?This is a very challenging problem as the system has both flat and highly dispersive bands,? says the theoretical physicist. ?Nonetheless, we managed to show that the ground state of the system in the absence of a field decouples into a product of the ground state of graphene and the ground state of twisted bilayer graphene,? a property that has subsequently been confirmed by experiments. Their results further establish the dominance of insulating and semi-metallic phases in the presence of an electric field which are unique to the trilayer system, i.e., are not realized in twisted bilayer graphene. ?We are able to use our resulting phase diagram for the correlated normal states to constrain the form of the superconductor,? says Scheurer. ?Among other aspects, the resulting two superconducting candidate states we get are consistent with the unexpected stability of the superconductor in magnetic field seen in experiment.?

The relevance of the findings for the physics of twisted trilayer graphene is further attested by a subsequent collaboration with the group of Abhay Pasupathy from Columbia university. In a recent paper in Science, they report scanning tunneling microscopy (STM) data on this system. ?We show that the measured tunneling spectra exhibit significant interaction effects that can be qualitatively captured by the numerics of our work,? says Mathias Scheurer.

Male baby pregnancies are more likely to result in complications, possibly because they grow faster in the womb and require more nutrients and oxygen than supplied by the mother through the placenta ? the temporary organ that attaches to the wall of the uterus during pregnancy to help the fetus grow and develop.

Now scientists have discovered the sex of a fetus can affect how well the placenta actually functions along with factors such as diet-induced maternal obesity and stress, after studying pregnancies in lab mice.

Around one in 10 women are affected by pregnancy disorders, such as fetal growth restriction and pre-eclampsia ? a life-threatening high blood pressure condition for both mother and child ? but these can be difficult to predict and treat.

The new research suggests that designing individual treatment plans and encouraging pregnant women to make lifestyle changes based upon the sex of their unborn babies could have lifelong health benefits for their children ? and spell the end of the tradition of waiting until the baby is born to find out whether it is a boy or a girl.

?Often parents don?t want to know the baby?s sex because they want it to be a surprise. But actually knowing the sex would help to identify whether a pregnancy may be at greater risk than another because we know that some conditions of pregnancy such as pre-eclampsia and fetal growth restriction can be more prevalent in women that carry male babies than females,? said Dr Amanda Sferruzzi-Perri, a Fellow of St John?s College, who carried out the research with her team from the University of Cambridge Department of Physiology Development and Neuroscience at the Centre for Trophoblast Research.

?We don?t quite know 100 per cent why that is but it might be related to the fact that male babies grow faster within the womb. So it might be that their demands for nutrients and oxygen supplied from the mother through the placenta can easily become limited, so the male baby may not be receiving all that it really wants and needs to grow to its full capacity. It may be that its resilience against stresses or poor conditions in pregnancy may be lower than say, for females, who have got less requirements.?

Dr Sferruzzi-Perri?s research team has revealed its findings in a paper published in the journal Biology of Reproduction. A second study written in collaboration with fellow researchers in institutions across the world, which demonstrates how diet-induced maternal obesity changes the structures of the placenta and affects baby growth, including causing alterations based upon the fetuses? sex, appears in the journal Acta Physiologica.

Fetal growth depends upon placenta function, which requires energy from mitochondria ? special compartments in our cells that help turn the energy taken from food into energy that cells can use as fuel. The researchers have found that in a normal mouse pregnancy, the placenta responds differently depending on whether it?s supporting a female or male fetus, and functions better with the lightest female fetuses, than for the heaviest male fetuses.

?The placenta has an amazing skill in changing how it forms and how it functions,? said Dr Sferruzzi-Perri. ?This can be seen at multiple levels from the way in which the cells form in the placenta, it?s genes and proteins, even its mitochondria. These can all change in response to different types of cues, whether it?s in a mother that has been eating a sugary, fatty diet, or in a pregnancy where there is rivalry between the siblings when there?s more than one baby, but what I think is probably the most novel aspect is that the way the placenta does adapt seems to depend on whether the baby?s female or male.?

The studies show the importance of designing sex-specific therapies for placental insufficiency and fetal growth abnormalities, as well as personalised lifestyle interventions or therapies for obese pregnant women.

?The data that we?ve been generating in the lab has really shown us that we have to consider the sex of the baby when monitoring a pregnancy,? said Dr Sferruzzi-Perri. ?Therefore maybe treatments should be tailored based on the sex of the baby.?

Scientists already know that male and female risk for diseases such as type two diabetes and cardiovascular disease is dependent on sex ? males tend to have a higher incidence of both. ?Whether part of this originates from early life exposures is quite a key area that people like myself and others are really eager to try and address. One way we are doing this is in mice using environmental conditions, such as diet-induced obesity, which is really widespread globally, including even in the developing world where urbanisation is meaning that women are having more access to sugary, fatty diets than what they have been before,? said Dr Sferruzzi-Perri.

In the UK more than half of pregnant women have a body mass index above the normal healthy range. This is concerning as maternal obesity increases the risk of pregnancy complications, such as miscarriage, gestational diabetes, pre-eclampsia and stillbirth. Abnormal birth weights can also lead to neurodevelopmental issues and immune disorders in childhood, and to obesity, diabetes and heart disease in adulthood.

?We?re now building more and more evidence of what to measure in the mum in pregnancy like her starting body mass index, her growth, her gestational weight, but also considering fetal sex. Routinely, clinicians do consider sex when they?re looking at ultrasound images, because sex is an important determinant of how the fetus is growing. However, we?ve not really understood before how that might be determined; how that might be interacting with the environment of the mother or the way in which the pregnancy is occurring. So our studies are giving more information to the clinician to provide more informed decisions about how to manage that pregnancy.

?It might be that a woman who has a male baby might need to adopt different lifestyle conditions, than a woman that is carrying a female.?

Currently, there are very few ways to treat women with pregnancy complications other than bed rest, dietary advice, or delivering the baby early, because of the risks of drugs passing over the placenta and affecting individual organs of the fetus, which are very sensitive. Scientists are now trying to think of ways to design treatments that just go to the placenta.

?These therapies would increase the way in which the placenta functions, how it develops, even how at the level of its mitochondria, it produces energy to support fetal growth,? said Dr Sferruzzi-Perri. ?A lot of this groundwork is reinforcing to us that the placenta is fundamental for healthy pregnancy outcome, and that fetal sex is important. But by looking at the different sort of gene, protein and cellular mechanisms, we might identify targets that could be like biomarkers and specifically targeted in the placenta to improve outcomes for mothers and their babies.?

Selection Bias Analysis Supports Dose-Response Relationship between Level of American Football Play and Chronic Traumatic Encephalopathy Diagnosis. American Journal of Epidemiology, 2022; DOI: 10.1093/aje/kwac075

Now a novel study by researchers from BUSM and Boston University School of Public Health (BUSPH) provides further evidence suggesting a dose-response relationship between football and CTE even after adjusting for selection bias.

CTE is a progressive brain disease associated with repetitive head impacts. It has been diagnosed after death in American football players and other contact sport athletes as well as members of the armed services and victims of physical abuse.

?The validity of research involving CTE has been questioned due to the inherent bias in the selection process of brain donation studies. A prior BUSM study that identified a strong dose-response relationship between years of football played and likelihood of developing CTE, which supports a cause-effect relationship between repeated head impacts and CTE, has been ignored by some sports organizations due to claims of selection bias. To respond to such criticism, there is a significant need to adjust for it in the analytical phases of these studies,? explains corresponding author Yorghos Tripodis, PhD, professor of biostatistics at BUSPH.

Utilizing data from Veterans Affairs-Boston University-Concussion Legacy Foundation (VA-BU-CLF) Brain Bank, the researchers undertook a new study to analyze 290 male former football players to further clarify the dose-response relationship. After developing multiple statistical models to adjust for selection bias, college and professional football players respectively were found to have a 2.38 and 2.47 times higher risk of being diagnosed with CTE relative to high school players.

?Strikingly, these risk ratios were actually heightened after accounting for selection bias. This is in stark comparison to the previous critiques received as it suggests that prior data may actually underestimate risk of CTE in these populations,? said Tripodis.

The researchers also calculated the minimum cumulative incidence of CTE for different levels of play. They found the minimum possible cumulative incidence for professional players is one in 10, one in 250 for college football players, and 1 in 20,000 for high school players. However, they anticipate future studies will reveal the actual risk is many times higher for each level of play.

The authors end by prompting the need for further investigation into the type of participants that are under-represented in the brain bank leading to this selection bias and the need for additional recruitment efforts targeted at these populations to allow for additional study.

?Our new study provides further evidence to support a dose-response relationship between playing football and developing CTE. We hope the football community will accelerate reforms aimed at preventing CTE by limiting head impacts for players and that parents will use this information to make more informed choices about sports participation for their children,? he adds.

These findings appear online in the American Journal of Epidemiology.

Roku?s expansion into original content continues with news that the streaming platform and device maker will debut its first original feature-length film during the 2021 holiday season. In partnership with Lionsgate, Roku announced it will introduce a movie based on the Emmy-winning TV series ?Zoey?s Extraordinary Playlist,? which will air on its free streaming hub, The Roku Channel, later this year.

The new movie, ?Zoey?s Extraordinary Christmas,? will continue where the series left off following its season 2 finale and series cancellation, which had upset fans who had taken to social media in hopes of saving the show by finding it a new home. Roku, as it turns out, is that home ? not only will it air the new film, it will also make all 25 episodes of the series available to stream for free on The Roku Channel in the U.S. later this fall.

The feature film, meanwhile, begins production this month in Vancouver and will see a number of actors reprising their roles, including Jane Levy, Skylar Astin, Alex Newell, John Clarence Stewart, Andrew Leeds, Alice Lee, Michael Thomas Grant, Kapil Talwalkar, David St. Louis, Mary Steenburgen, Peter Gallagher and Bernadette Peters. Details of the film?s plot are still limited, however, noting only that it will be a continuation of Zoey?s journey as she ?navigates work, family, love and everything in between.?

While a significant move for fans who simply wanted more of their favorite show, it?s a bigger step forward for Roku and its original content strategy. The company had already tested consumers? appetite for original series by acquiring the content catalog from the short-lived streaming service Quibi earlier this year, whose shows then launched to The Roku Channel as the company?s first set of Roku Originals. The debut lineup included 30 titles, but Roku is continuing to roll out more shows as the year progresses. Last month, for instance, it released 23 more of those shows to its collection.  The company also recently decided to reinvest in at least one previously Quibi-owned series by greenlighting a second season of ?Most Dangerous Game.?

But until now, Roku has not invested in full-length movies.

Movies make a good fit for The Roku Channel, of course, as the free streaming is supported by advertising ? and movies, due to their length, offer more ad slots to be filled. In fact, The Roku Channel got its start as a free movies hub, before later expanding to include TV shows and other types of content, like news and sports.

So far, Roku?s original programming seems to be connecting with viewers. The company says its top 5 streamed TV shows this summer (May 20-July 18) were all Roku Originals.

The new film is produced by Lionsgate in association with the Tannenbaum Company, Feigco Entertainment and Universal Music Group?s Polygram Entertainment and Zihuatenejo Productions. The series creator Austin Winsberg will write and executive produce. Richard Shepard, who filmed the show?s pilot, will direct. Kim Tannenbaum, Eric Tannenbaum, Paul Feig, David Blackman and Daniel Inkeles will also serve as executive producers.

The movie will air on The Roku Channel during the holidays in the U.S., Canada and the U.K.
